Kristen played Bella mentally stronger. In the books, we see that as the reader, but Kristen was able to bring that out of the book. She understands Bella's character and she found a way to bring her inner strength outward.
Rob played Edward more intense. In the books, Edward is very calm but gradually becomes more intense as all of these events happen around Edward and Bella. Since there are only so much time for a movie, Rob's Edward came off as more intense than the book's character. Whether this was intentional, I'm not quite sure. And a lot of Edward's selfless deeds aren't portrayed in the movie.
Taylor played Jacob, not as the victim, but lighter than how Jacob is in the books. In the books, Jacob becomes two sided. He becomes more aggressive and is more impeding attitude. We hardly witnessed any of Jacob's "post-transformation" side. It was more noticeable in the book.
I like what they did to their characters to an extent and with an exception of an actor, but as you can see, there are a lot of differences, and it's hard to answer this question because of that. The minor characters are the only people I think, that are candidates of this question because their characters weren't expanded on too much.
I would have to say Ashley Greene played Alice the best, but only because she had little room to improvise; little room to be the character.
