“Lazarus Rising” is a reference to the biblical story of Lazarus of Bethany. Lazarus was resurrected from the grave by Jesus. (John 11:25–26 KJV). In that story, Jesus states “I am the resurrection, and the life: he that believeth in me, though he were dead, yet shall he live: And whosoever liveth and believeth in me shall never die.”
So, basically Dean is compared to Lazarus, because he was brought back from hell by Castiel.
