Peter Parker is a lonely, unpopular, and nerdy high schooler. His parents were killed when he was a small child. His Aunt May and Uncle Ben Parker adopted and raised him as their own child. The love of Peter's life is Mary Jane Watson, the most popular girl in high school.

While on a class field trip to the museum, Peter is bit by a radioactive spider. The insect's venom gives Peter all sorts of unusual powers: the ability to climb walls, shoot webs, superhuman strength, and a sort of "spider sense" that tingles whenever Peter is near danger. Peter creates a blue and red costume for himself and dons the name "Spider-man". Peter uses his new powers to win some easy cash in a wrestling tournament. When the fight promoters refuse to pay Peter his winnings, Peter allows a thief to get away with the promoter's money.

When Peter arrives home later that evening, he finds out that his beloved Uncle Ben was murdered by a burglar. Enraged, Peter puts his Spider-Man costume back on and sets out to find the criminal who murdered the man who raised him as his own son.

Spider-Man tracks the criminal to a warehouse and subdues him. To Peter's shock, the villain is the same thief who robbed the prize money at the wrestling match earlier that evening. If Peter would have only captured the thief during the robbery, the death of his Uncle Ben could have been prevented.

Feeling extreme guilt, Peter decides to become a full-time crime fighter, in an attempt to make up for the great tragedy that he feels responsible for. As Peter's Uncle Ben once told him, "With great power comes great responsibility". As Spider-Man, Peter Parker uses his newfound gifts to protect the innocent from crime and havoc whenever possible.

Spider-Man's main enemies are The Green Goblin, Sandman, Electro, The Vulture, Rhino, Mysterio, Venom, Carnage, Doctor Octopus, and The Lizard.
