Buffy's Death: I am absolutely positive that this was tramatic for Willow and, until Seeing Red, I believe this was the event that probabaly shaped her character the most going into season six. My main reason for not picking this is because she had a strong support system to fall back on - most importantly Tara. Her best friend, who she loved, died but she still had who I consider to be the love of her life with her.
Xander ignoring her - To me this is just the normal sort of thing that happens to people as they grow up. Whoever you fancy will naturally not notice you or take forever to see you. It's an unwritten law.
Having Oz leave - This hurt. A lot. Oz was her first love and he will always have a piece of her heart but he didn't die. Oz is alive and and split with saying how much they loved one another. There was a degree of closure with these two.
Going dark - I consider Dark Willow to be the manifestation of Tara's death. Tara's death is what caused Dark Willow and one cannot be viewed without the other. Going dark did take a giant toll on her self confidence but it was the loss of Tara, her counter balance, that threw her hammer out of wack completely. Willow would never have gone dark without the pain of losing Tara and she also had to deal with it being a mortal death that she could not fix no matter how dark she went (there is a series of novels called Dark Willow, it's an AU series that explores Dark Willow searching for a way to bring Tara back, it's decent but not amazing).
Simply stated, Tara's death was the removal of an unresolved relationship and the loss of her biggest support system, it sent her into a tailspin and changed everything for her character.
